- It has been reported that the extent of maleimide ring opening by hydrolysis varies depending on the linker structure in the conjugation pattern via cysteine residues. In this study, the amount of linker hydrolysis was quantified by LC-MS/MS analysis, revealing that the extent of hydrolysis differed based on sample storage conditions.
- Linker hydrolysis affected chromatographic behavior, particularly influencing reversed-phase chromatography and cation exchange chromatography.
- Molecular weight evaluation and post-translational modification assessment by mass spectrometry indicated that the impact of factors other than linker hydrolysis on the chromatographic assays was limited, clarifying that linker hydrolysis primarily affects the analytical results.
